PMID: 12865942 Here we delineate that it acts as a cells self compensatory response against the exposure with BPA. Here we observed the role of autophagy againsts the BPA induced oxidative damage. We had treated the neuronal cell with the antioxidant enzymes like SOD( Superoxide dismutase) as well as NAC (N-acetylcysteine) prior to BPA exposure. There have been several literatures regarding BPA which suggests about NAC protective response against BPA induced cognitive dysfunction and oxidative stress in rats. PMID: 21440025 Thus both the NAC as well as SOD acts in reducing the oxidative stress as well the generation of ROS. Through the immunoblotting results it was observed that LC3II expression was significantly reduced due to the prior treatment with the antioxidant enzymes before BPA exposure in neuronal cells. Further in order to validate autophagy as a self compensatory response against the BPA induced neurotoxicity, we performed an experiment in which we had treated the neuronal cells with rapamycin as well as with Wortmannin along with BPA and we found that ROS generation decreases after rapamycin but it was enhanced after the Wortmannin treatment. Suggesting the protective efficacy of autophagy. Next we used Beclin siRNA along with BPA and we found that ROS generation was increased after the knock down with Beclin siRNA .Thus it was concluded that rapamycin suppressed BPA induced ROS generation whereas Wortmannin as well as beclin siRNA enhances the ROS generation. Hence it may be concluded that autophagy acts as a self –compensatory response against the BPA induced neurotoxicity as well as the oxidative damage.
