The ability to be able to regenerate bone tissue by tissue engineering may be the first method of regeneration to be successful. Researchers are constantly looking for solutions to reduce the amount of complications that accompany dentistry and/or orthopedic treatment of congenital anomalies, trauma, periodontitis, and other diseases (Alsberg et al. 2001). Though the common treatments may include autograph tissue (tissue that was taken from one part of the body and placed in another part of the same body) or artificial teeth, there are many complications that are associated with these types of treatments (Xiong 2002). Some of the manufacturing process for scaffolds include fiber bondng, solvent casting, particulate leaching, melt molding, emulsion freeze drying, precise extrusion, and fused deposition modeling (Xiong et al. 2002). There are two types of scaffolding techniques that will be covered: regular scaffolding and bilayer scaffolds.
Regular scaffolds in one experiment were manufactured using a new technique named low-temperature deposition (LTD) manufacturing using Poly L-lactic acid (PLLA) and tricalcium phosphate (TCP) as components for the scaffold. The new process used computer-aided manufacturing to develop a layer-by-layer process of solid freedom fabrication (Xiong et al. 2002). The histological analysis data reported that after implantation of the PLLA/TCP composite scaffold, they were degraded completely after 24 weeks with no trace of aseptic inflammation and it was concluded that the composites were a better choice when used together than when used alone. The porosity of the scaffolds were up to 89.6%, which controlled interconnective macrocellular and microcellular morphology which aided tissue regeneration since they were very similar to human spongy bones (Xiong et al. 2002). The scaffolds also proved that they were biocompatible, had good bone conductivity, and had the appropriate amount of biodegradation in vivo for bone repair. Altogether, the LTD manufactured scaffolds were suitable tissue regeneration scaffolds for bone tissue engineering.
Bilayer scaffolds are unique in that each layer makes it more feasible to create separate and specific biological environments using varied structural and chemical properties. To manufacture these scaffolds, a synthetic polymer material is used to provide a template for mechanical strength and a hydrogel membrane is used to provide cell support in the scaffold. In an experiment developed Hydroxyapatite (HAp), a mineral form of calcium, was used in nano-particle form as a composite for the bilayer scaffolds (Zhou 2011). HAp was chosen since it provides exceptional biocompatibility with soft tissue: skin, gums, muscle, etc (Maheshwari et al. 2014). The pore size in the bilayer scaffolds were adequate enough to allow the cells to move or adhere to the scaffolds which allowed for rapid multiplication of cells.
