Ketoprofen was obtained as ex gratis from Ciron Drugs & Pharmaceuticals Pvt. Ltd, Mumbai, India. Phospholipon 90H (Hydrogenated soyphosphatidylcholine, HSPC) sample was obtained as a generous gift sample from Lipoid, Germany. Mannitol, compressol, erythritol, pearlitol SD 200 (Spray dried mannitol), pearlitol DC 300 (Directly compressible mannitol) and directly compressible lactose 22 were generously provided as gift samples by Dr. Reddy’s laboratories, India. Cholesterol was supplied by Hi-Media, India. All other chemicals utilized throughout the study were of analytical grade and solvents were of high performance liquid chromatography (HPLC) grade. Fresh double distilled water was used throughout the study.
Briefly high performance liquid chromatography (HPLC; LC-10 AT, isocratic Shimadzu, Kyoto, Japan) furnished with SPD-10 AVP UV spectrophotometric detector (sensitivity of 0.005 absorbance units full scale; AUFS) and Phenomenex C18 column (5 m, 4.6mm×250mm) was used to analyse ketoprofen samples at detection wavelength of 256 nm. The mobile phase composed of acetonitrile and 20mM potassium dihydrogen orthophosphate buffer (70:30 % v/v; pH adjusted to 3.0 with orthophosphoric acid) delivered at a flow rate of 1 mL/min. The samples of ketoprofen (20 L) were injected into the column maintained at room temperature.
Preparation of proliposomes using different carriers
Ketoprofen loaded proliposomes were prepared employing different carriers such as mannitol, pearlitol SD 200, pearlitol DC 300, compressol, erythritol and directly compressible lactose of 1g per 1mM of total lipid mixture using 1:1 molar lipid ratio of HSPC and cholesterol as lipid mixture (250 µM) by solvent evaporation method (22,23). The drug (25 mg), lipid (HSPC) and cholesterol were dissolved in 10 ml of chloroform and absolute ethanol as solvent mixture (1:1) and then the solvent was allowed to evaporate under vacuum using rotary flash evaporator (Laborota 4000, Heidolph, Germany) at 37oC.
